ly be excited from 300-380 nm. Phos Yellow 2? shows a much stronger phosphorescence at ≈ 540 nm than Phos Yellow? in air equilibrated glycerol, and similar to Phos Yellow? has a FWHM of ≈ 100 nm. The Phosphorescence tails into the yellow spectral region, which accounts for its unique yellow phosphorescence. The lifetime in air equilibrated glycerol is ≈ 113 ?S.
Phos Yellow 2? is sold dry and is simply reconstituted by adding MeOH, EtOH or another suitable solvent. One vial of this probe will typically allow for 10 measurements to be undertaken:? 50 ?L of MeOH is typically added to each dry vial. Subsequently, ten 5 ?L aliquots diluted to 3 cm

(cuvette), will typically give an optical density > 0.1 at 320 nm (in spectroscopic MeOH).
For optimum performance, probe vials should be stored at -2 to -8 C when not being used. Probe solutions should be used after dilution.
