Experiment Name:
Antibody titers to ST-CF of different IgG isotypes.
Experiment Background:
The relative levels of specific IgGl and IgG2A induced by the different doses of ST-CF were used to give an indirect indication of the lymphocyte subsets involved.
Experimental Steps:
1. Serum samples obtained from the mice 5 weeks after the first immunization were analyzed in two fold dilutions on
ELISA
platescoated with ST-CF (100 ?l per well; 5 ,ug/ml). 2. The plates wereincubated for 2 h at room temperature, and, after washing, theplates received peroxidase-labeled goat antimouse immunoglobulinG (IgG), IgG2A (1/1,000), or IgGl (1/2,000) (Kidlington, Oxford, United Kingdom). 3. Antibody titers toST-CF of different isotypes are expressed as the dilutions thatgave an absorbency of 1 (IgGl) or 0.4 (IgG2A).4. The titers havebeen adjusted for different levels of specific IgG induced by thevaccines and are therefore presented as relative isotype titers.
