SFR Luciferase Reporter HEK293 Cell Line is derived from human embryonic Kidney, and stably express firefly luciferase reporter gene under the control of SFR response element. This cell line is an ideal cellular model for monitoring the activation of SFR response Pathway triggered by stimuli treatment, enforced gene expression and gene knockdown.
Principle:
Serum response factor (SRF) is a member of the highly conserved MADS box family of transcription factors which regulates the expression of immediate early genes. SRF binds to its specific promoter sequence (CArG box). SRF encodes a ubiquitous nuclear protein that stimulates both cell proliferation and differentiation. SRF binds to the serum response element (SRE) in the promoter region of target genes, regulating the activity of many immediate-early genes, for example c-fos and actin, thereby participates in cell cycle regulation, apoptosis, cell growth, and cell differentiation, and cytoskeletal regulation.? This gene is the downstream target of many pathways; for example, SRF co-activators are activated by the MAPK pathway through the ternary complex factors (TCFs) in response to mitogenic and stress stimuli. In addition, SRF specific co-activators the MAL/MRTF proteins are released from a repressive complex with G-actin upon induced changes in actin dynamics.
