ELISA
based assay measures the loss of biotinylated poly (ADP-ribose) from immobilized histones in a 96-well format due to PARG activity. The addition of Strep-HRP (biotin-binding protein) and a colorimetric HRP substrate yields relative absorbance that correlates with PARG activity.
