Tribbles homolog 3 (TRB-3) is a pseudokinase, shown to act as a potent negative regulator of PPARγ, a master regulator of
ADI
pocyte differentiation, and to control
ADI
pogen
esis. It disrupts insulin signaling by binding directly to Akt kinases and blocking their activation. It interacts also with the NF-κB transactivator p65 RELA. May play a role in programmed neuronal cell death but does not appear to affect non-neuronal cells.
